Extraction of good quality DNA is the most essential requirement for molecular analysis of any species. Five protocols for DNA extraction were compared to assess their effects on amount of DNA recovered, its purity and efficacy in PCR amplification using RAPD, ISSR and SSR markers in two species of Dendrocalamus asper. and D. hamiltonii. Leaf tissues from in vitro cultures, tissue culture raised plants in green house,and young and mature leaves from field grown plants of the two species were subjected to five DNA extraction methods using the protocols. Significant differences in DNA quantity and quality between samples and protocols were observed. Leaves of tissue culture raised plants growing in the green house gave best DNA yield (5681, 5015 ng/ μl)followed by in vitro shoot cultures (4645, 4379 ng/ μl), and young (2720, 1801 ng/ μl) and mature leaves (1800, 1368 ng/ μl) from field grown plants of D. hamiltonii and D. asper respectively.The concentration of DNA obtained was more for D. hamiltonii samples than the corresponding D. asper tissues.Keywords

Commiphora wightii (Arn.) Bhandari, a balsamiferous woody shrub has remained an important component of many medicinal formulations since 2000 B.C. for the oleo-gum resin Guggulsterone, E and Z, the two important isomers, confer lipid- and cholesterol-lowering, and anti-cancerous properties to guggul gum. Tapping of oleo-gum resin is usually done by local and tribal people adopting unscientific methods which generally results in the deathof plants. Rajasthan, Gujarat and Haryana, three hot spot diversity rich Indian states were covered during survey, exploration and collection assignment of guggul for analyzing the extent of genetic variability as well as their socio-economic importance. A total of 78 accessions, both wild and cultivated, were collected from surveyed regions and 52 of them could be established in the green house by ischolar_mained cutting method. Attempts were also made to establish the species in vitro however limited success could be achieved. During the survey, sizable variations were recorded in morphological and geographical traits in different accessions. Modern as well as traditional potential applicability of this rocky desert medicinal plant was explored and investigated during this mission.Keywords

A diverse array of growth regulators interact at the cellular level to produce physiological and morphological effects on plant growth, morphology and yield. The five conventional growth regulators viz. auxins, cytokinins, gibberellins, abscisic acid and ethylene are being used in plant cell, tissue and organ cultures for decades, while many of them, like non-purine cytokinins, polyamines, jasmonates, brassinosteroids, oligosaccharides, sterols, phosphoinositosides, salicylic acid and systemins, have recently been discovered and tested for their effects in vitro. However, many have not yet been examined for their effects on in vitro growth and development of plants. In this review, we attempted to summarize the progress that has been made over the past two decades towards understanding the role of non conventional PGRs in plant growth and development.Keywords
